Survival quantitative trait locus fine mapping by measuring and testing for Hardy-Weinberg and linkage disequilibrium.

I show that fine-scale localization of a survival-related locus can be accomplished on the basis of deviations from Hardy-Weinberg equilibrium and linkage disequilibrium at closely linked marker loci. The method is based on chi(2)-tests and they can be performed for age-specific samples of alive (or dead) individuals, as for combined samples of alive and dead individuals.

A Bayesian characterization of Hardy-Weinberg disequilibrium.

A Bayesian method for determining if there are large departures from independence between pairs of alleles at a locus, Hardy-Weinberg equilibrium (HWE), is presented. We endorse the view that a population will never be exactly in HWE and that there will be occasions when there is a need for an alternative to the usual hypothesis-testing setting. Bayesian methods provide such an alternative, and...

Lee Responds to “Testing for Hardy-Weinberg Disequilibrium”

I appreciate Weinberg and Morris’ thoughtful commentary (1) on my paper (2). In their article, they put my work under the perspective of gene mapping in the postgenomic era. I share the same view with them that the method proposed in my paper amounts to a tree-shaking approach to harvesting the high-hanging fruit (a low-cost approach to generating hypotheses aimed at localizing diseasesusceptib...

A comparison of linkage disequilibrium measures for fine-scale mapping.

Linkage mapping generally localizes disease genes to 1- to 2-cM regions of chromosomes. In theory, further refinement of location can be achieved by population-based studies of linkage disequilibrium between disease locus alleles and alleles at adjacent markers. One approach to localization, dubbed simple disequilibrium mapping, is to determine the relative location of the disease locus by plot...

Disequilibrium likelihoods for fine-scale mapping of a rare allele.

Genetic linkage studies based on pedigree data have limited resolution, because of the relatively small number of segregations. Disequilibrium mapping, which uses population associations to infer the location of a disease mutation, provides one possible strategy for narrowing the candidate region. The coalescent process provides a model for the ancestry of a sample of disease alleles, and recom...
